La historia versa sobre un travieso perro de raza Gran Danés que vive con la familia Winslow. Este perro apareció después en varios episodios animados de 'Heathcliff' (Isidoro) y 'Garfield and Friends'. La película mostrará cómo Marmaduke se encuentra tras mudarse de domicilio en medio de una lucha por el territorio entre chuchos callejeros y perros con Pedigree, además de que intentará conquistar a la perra de sus sueños.
